Thankyou so much for offering! Well the DNA technology section is bit lacking in my notes, specifically in vivo and in vitro gene cloning and locating and sequencing genes

Ive been doing some past papers and I dont really get restriction mapping?

Is there anyone who has anything or can explain restriction mapping to me??
iv read over the revision guide and thought i understood it (kindof) but then i answered a question on it didnt get it and didnt have a clue what to write!
any help would be much appreciated!

Please may someone briefly explain muscle contraction? Getting so confused right now would be much appreciated, thanks!

Those essays are from that synoptic essay book, they're so advanced and long, I could neeever write anything like them! :emo:

The action potential reaches the muscle and travels down the t-tubules, this causes Ca2+ ions to diffuse into the sarcoplasm, they causes tropomyosin to move away from the actin, unlocking the binding sites. There is an ADP molecule on the myosin head which means it can bind to the actin to form cross bridges, the myosin head changes its angle pulling the actin along, the ADP is also then released. ATP attaches to the myosin head causing it to detach and ATPase (activated by Ca2+) hydrolyses ATP so myosin can return to its original position. Due to hydrolysis of ATP, myosin head has an ADP on its head again so it can reattach to actin's next binding site. This is repeated while stimulation continues

Yeah I just realised as I was reading them haha, I could never write something like this, I struggle just picking out some facts cause there's so much to write! Plus I'm one of those people who panic in the exam and go blank -.-

I could upload you the 'writing the synoptic essay' book in e-book format if you like?

Can someone tell me the role of ATP in muscle contraction. I'm not sure if its used to break the actomyosin crossbridge or something else :/ Thanks

Aww thankyou so much! The notes are great and that powerpoint is brilliant. I was overwhelemed by the essays! Really detailed, I dont think I could write in such depth in exam conditions, do you think we have to write to that standard?

Anways, thanks a million and I wish you all the very best of luck for your exams!

i was just wondering that aswell, cause the author seems to have a little intro for every point he makes. Do we have to make any small intros like that, or do we just get straight to the point?

Id get straight to the point if it was me and spend time on the section that added more weight to my essay but agian it needs to be well written too. Id obviously link what I was saying though in terms of the topics, bit unsure tbh..and there really long! Im so scared im going to run out of time at the end of the exam

Having read them essays, its **** me up lol. If we're expected to write along them lines, then i dont stand a chance at all:/ I'm just hoping they dont have any questions that were on the biol4 paper yesterday, like the petroleum question lol

Thank you and thanks to the other person who answered. So i'm guessing:
    Iron - haemoglobin
    Calcium - muscle contraction/synapse
Nitrate - nitrogen cycle (could also mention ammonium & nitrite)
    Hydrogen - respiration (ETC)
    Potassium - resting potential/action potential
    Sodium - resting potential/action potential
Chloride - Cholera, cystic fibrosis (water potential)
    Phosphate - DNA?

I really want this essay to come up now, there's loads to write!
Hydrogen ion during photosynthesis is plants as it is produced during the photolysis of water and combines with electrons and then reduces NADP.
Significance, r.NADP used in LIS to reduce GP so if H+ wasn't produced in LDS this could not happen and photosynthesis may cease.

P.s. Why do people think it will be an ion essay?

Could someone explain the sliding filament theory please????? I have no clue
ill explain it from the nerve impulse.

-Action potential arrives at muscle- impulse travels through t tubules.
-this causes calcium to be released into the sarcolemma from the sarcoplasmic reticulum
-calcium binds to tropinin causing it to move the tropomyosin (imagine tropomyosin as a ribbon covering the binding sites)
-actin binding site is now exposed and myosin(which has an ADP molecule attatched) attatches to binding -site on actin fillament(cross bridges form)
-myosin head pulls backwards moving along the actin filament- ADP is released
A-TP attaches to myosin head caising it to detach from actin filament
-hydrolysis of ATP recocks myosin head

as this happens the sarcomere gets shorter

    hope this has helped
cycles is repated further along actin filament.

Anyone got any calculated guesses for the essay? Guessing there is no chance of cycles or DNA because they were on last year?

Gutted because I would have loved cycles!

Does anyone fully understand the section in unit5 on locating and sequencing genes?

For anyone using the nelson thornes book, could you help me?

On page 267 of the book it says 'before we can make a specific probe we need to know the sequence of nucleotides in the particular gene that we are trying to locate'

So does page 267, 268 and 269 just cover the sequencing information or does it also cover the making of the probe too?

im really confused with all these steps lol
