Although I voted for Aliens, there is some body of evidence that it's actually a war film rather than a scifi film - and James Cameron seemed to think it was more specifically a Vietnam War film.
I would have gone with Bladerunner for being the sine qua non of Cyberpunk; but it lost out for me due to the differing cuts and endings. -

In terms of scope and demonstrating what the genre's capable of, 2001 is pretty much the king. Gets a lot of criticism for flat characters and slow pacing though (even though both are intentional). Considering story and character, you'd have to go for Blade Runner. For pure entertainment/the childish wonder that big, blockbuster, event cinema is supposed to evoke, it's unquestionably Star Wars.

Terminator 2. Just.. Awesome. My 13 year old self was blown away when I spent time watching the Terminator films, I just really wanted to get into that one and I did, never looked back, my favourite film! -

I'm not going to vote because I haven't seen all the films, but surely Metropolis deserves to be on a 'sci-fi greatest' list?
From that list I went for The Matrix, though 2001 and Star Wars are also good. For modern sci-fi I liked Minority Report.
