Open with a clear statement of what the letter is about eg This is an application for the post of XYZ, advertised in ABC

Then a few sentences on why you want to work in that industry/volunteer.

Then a few sentences on why you are choosing that company/charity.

Then highlight your one or two most relevant skills.

Then end with any technical details they are asking for (languages, visas etc) and interview availability if relevant, then thanks.

Don't waffle. Don't resort to a great long ramble about how super-duper you are. Don't get smarmy about how wonderful the company/charity is. Keep is short, polite and relevant.
