The gene in question produces a GFP. The gene to be cloned is transplanted into the centre of the GFP gene. Any bacterial cell that has taken up the plasmid with the gene that is to be cloned will not be able to produce GFP and will not fluoresce.
page253

Is this a mistake in the NT textbook?
Surely, if the host cell has taken up the plasmid with gene to be cloned (which has been transplanted into the GFP gene), then GFP protein should be produced?

I think what they mean is if the gene that you want cloned is inserted into the GFP gene (thereby interrupting it), any bacteria that is cloned from it will not fluoresce after as a result of this interruption.

Please God let the essay be on ATP as that's the only one I'll be preparing for...

I still need to go over a bunch of unit 2 stuff, I hate that this exam is synoptic :frown:
Doing really bad in past papers and dreading the essay.

Right now I'm just doing questions from previous spec. papers, I only have one more current spec. paper to do, but I wanted to do one on the day. Tomorrow I'll also be looking over my notes from BIOL1/BIOL2/BIOL4 again, followed by re-reading essay plans, then about an hour before go back read over unit 5 topics. Absolutely praying that the essay title is something on protein shapes (this is what my teacher predicts, because proteins pop up everywhere)!

Strangely, I'm not nervous about this exam, although that is subject to change. What is everyone else feeling about it? Good? Bad?

At least it's only the essay that's synoptic. :frown:
But yeah, your second line applies to me too.

I need a B overall for uni. I've been told to aim for a low B in this exam, I'm not sure though.
How about yourself?

It's 40 minutes for the essay leaving you 95 minutes for the questions.

I'd advise 5 minutes (10 minutes maximum) for the plan.

Out of curiosity, do you gain marks for a plan?
Guys, are TAQ polymerase and DNA both used in PCR as in, will the MS accept both?

I thought the whole paper was synoptic 😕 I've done past papers where some of the answers have been purely unit 2 stuff even though what I said could have been a valid answer :frown:

Yes you do because it shows you intended on talking about it but never got around to it, so, you knew what you were talking about.

Here's hoping the essay choices are on ATP, enzymes or ions, I swear if I knew an examiner personally I'd give them my life savings right now just to know those essay titles!!
