At temperatures above 40 C the shape of the active site alters and so the enzyme is unable to fit with a substrate. This means that the enzyme is denatured and the rate of reaction decreases rapidly.
Each enzyme also had an optimum pH. Most enzymes function within a range of 5-9 with the optimum being 7.
However rate of reaction also increases with temperature, so it's important to find the right balance.

For example, intestinal enzymes have an optimum pH of about 7.5. Enzymes in the stomach have an optimum pH of about 2.
