You isolate the chloroplasts, eg you just want chloroplasts nothing else so you can test them. The medium helps you do this from solid plant material i.e in the centrifuge.

The water bath keeps your experiment at a set temperature.

my teacher gave us these questions to prepare us for our ISA

    isolate it from what? thanks

also why is the equipment pre-cooled before the experiment?

and why do you use ICE in the water bath for the chloroplast?

why is testing on plants an advantage?

    thanks!!!

The rest of the leaf, nuclei, cell wall, membrane, fibres....

I'm not sure about the temperature but I think it is to do with preserving the chloroplasts, as they will start to break down.

Ice keeps things colder than water as more more energy is needed to change the state of ice (i.e for it to melt) than to heat water.

Plants are obviously an advantage because they have chloroplasts, also there are no ethica considerations... Not sure about that to be honest.

    Hope this helped
