In PCR will there be a leading strand and a lagging strand? I know the DNA strands will be denatured, but does this make any difference?

Just a random question stemming from my own needless curiosity!

There is no lagging strand

The DNA is completley denatured by heat, rather than gradually unwound by helicase.

Also, having a lagging strand would require many pre-synthesised primers or a heat stable primase, both of which would be counter-productive in molecular biology techniques.

you certainly did more than me! I didn't know about the lagging strand stuff until uni.... DNA is not that great. Proteins and signal transudction are much better.

Nooooooo I've had enough of signal cascades to last me a lifetime!! DNA is much more fun, especially multi-gene segment rearrangements such as is the case with generating immunoglobulin diversity! Now that is FUNKY! /geek :p:
