    What would be the most appropriate buffer for maintaining the pH at 8.5 during an enzyme assay of a cell-free extract at 250C?

    I'm guessing bumping granules won't help?

    What are the chemical reactants, and the chemical products in the reaction? Once you've considered this and how acid/alkali the products are, you can select a buffer than will maintain the more alkali pH of +8.5 (it won't remain exact of course).

    Bumping granules are used to stop boiling over. They are unreactive, like Tungsten or Inulin.
