At low temperatures enzyme molecules are inactive but undamaged and the rate of enzyme activity remains low. The temperature at which most enzyme-substrate activity occurs is at around 35-40 C. The temperate an enzyme works best at is called it's optimum temperature.

At temperatures above 40 C the shape of the active site alters and so the enzyme is unable to fit with a substrate. This means that the enzyme is denatured and the rate of reaction decreases rapidly.

Each enzyme also had an optimum pH. Most enzymes function within a range of 5-9 with the optimum being 7.

if the temperature is too high, the enzyme may denature (as the active site changes shape), and so stop working...
However rate of reaction also increases with temperature, so it's important to find the right balance.

If the pH changes this can also change the rate of reaction, because enzymes have an optimum pH that they work best at
For example, intestinal enzymes have an optimum pH of about 7.5. Enzymes in the stomach have an optimum pH of about 2.
