Genetics - undergraduate level

I am struggling to understand genetics and whilst revising I have come up with a few questions that and I was wondering if someone could clarify please. I've seperated them into topics.

Gene mapping:
- What are restriction fragment length polymorphisms?
- I don't understand how physical mapping works and what yeast artificial chromosomes (YACs) do?

Chromatin Modification
- What are CpG islands? How come methylation of them causes inactivation?
- What is chromatin and what is heterochromatin and what's the difference between the two?

Genome Organisation
- Stupid question but does reverse transcriptase exist in humans?
