Why do the cells used in karotype have to be in metaphase?
Because the chromosomes become distinctly visible between the end of prophase and the start of metaphase. After that they start dividing so it wouldn't be possible to artificially arrange them into their homologous pairs.

This. Also, it would be eaiser to see the banding and centromere position (classic features of a karyotype) during the metaphase stage.

Well the stages of the cell cycle merge into one another, so at some point during prophase or at the very start of metaphase before the chromosomes line up along the centre, the photographs are taken
