ELISPOT

Can anyone verify my understanding of this technique? We're using it to assess for Th1 clones against a certain antigen. The plate has antibodies to IFN gamma attached to it. You add into the well Peripheral blood mononuclear cells (which includes monocytes and lymphocytes). You then add the antigen you wish to test. Is the idea now that the monocytes will process the antigen and present the fragments to the Th1 cells? Can monocytes do this or do they differentiate into macrophages?
