Is nightline or Samaritans a good way of learning about people and how to talk to people? Or would volunteering in a hospital or with St. John ambulance or something be better for that?

They're all the kind of thing that you would do when you have some existing skills, because you're helping people in need. If you have no communication and people skills you would be better to start off with something easier. A retail job or charity shop would be perfect

I've done a lot of customer service and am quite confident with the public now. Just want to take it a step further and progress to helping people in a crisis now. But I don't know which out of the ones I mentioned would be best for developing skills like this.
