Primer usage; temperature changes for H-bond breaking and strand annealing; only one DNA polymerase, which is thermostable, is used, whereas different organisms may use chemically different ones; lack of semi-conservative replication.

Stick to the primer+temperature+lack reasons though, the one about the enzyme may not be on markschemes
