Can someone help me with these questions?

-Which fractions should you pool together to recover the haemoglobin?
Is it 3, 4, and 5?

-Give two reasons why the fractions you have pooled should contain the haemoglobin protein.
Haemoglobin is larger and is eluted first as it does not enter the pores of the gel

-What is the void volume of this gel filtration?
Volume of buffer to elute molecule that doesnt enter pores
Vo = 4mL

I think Vo should be 2 ml.

Not an expert on gel filtration but i'm learning lol.

I think you should also talk about the absorption. Red sample will absorb at lower wavelengths while blue samples will absorb at higher wavelengths.

Thank you for replying! From the diagram in my lab book I thought it was the highest point of the large molecule. Do you know why red absorbs at lower wavelengths?
