So basically, what distinguishes the different between introns and exons? Is there a physical difference, or chemical (regarding the process of splicing)?

My guts tell me that due to the specificity of enzyme's active sites, they will only target the molecule they are complementary to, in this case whatever bit of gene they are programmed to cut.
