If you apply to a Cambridge college do you get an interview?

Also, is it possible if you do an open application and you get into one of the best colleges like Kings College or are colleges based on the subjects you want to study

Most colleges have a tutor in most subjects. So the choice of college is a finer detail.
If you make an open application, your application is automatically sent to the college with the fewest applicants (or maybe it's lowest applications:admission ratio). So probably not King's.
If you have a strong application but don't get in to your chosen college, your application may be pooled for different colleges to accept.

And yes, you get an interview at the college. They usually interview a high proportion of applicants.

There isn't such a thing like the "best college", and while some colleges like King's get a higher number of applicants per place on average, this is evened out by the pooling system (and apparently almost all people who get in think their college is the best :wink:) I suggest you read this https://www.undergraduate.study.cam.ac.uk/applying as a start, it basically answers all your questions.

As per the above replies but to add Cambridge interviews at least 70% of applicants and often nearer 80%.
