So there is this lady who is good friends with my sister who's fancied me since 2005 and she is 2 years younger then me. I'm in my 30th year. She walks past and flirts with me, I don't acknowledge her, try to ask her out, or even add or talk to her on Facebook.

HER GAMEPLAN

Her aunty said to her mom "as I get older, I have no choice but to accept your daughter".

"he will be put under pressure"

"He will not find anyone or take a long time".

So he has no choice but to marry your daughter.

They're playing games, going to come to my house all confident and manipulate me.

Her mom, aunty will say "she's a nice girl, very nice girl". "she's educated".

Why not?

HER BACK OUT "well you're still single and continue to be".

I would just avoid her at all cost. Ask your sister to not invite her when you are around or to at least let you know ahead of time if she is coming over. Clearly she’s hoping with all the advances she is making towards you would eventually wear you down and make you cave in to her wishes.
