I got flagged up by Scribbr and Unicheck for using a quote from the bible in my English A Level Coursework. Will I be penalised?

My understanding is that it's not that you use someone else's work but how you use it. Copying someone else's work, without referencing it, is plagiarism - which you should know about by "A" level. Which reference system does your institution use?

Check your course specification for details.

I don't know how WJEC checks for similarity actually. I didn't cite the bible reference as it was a creative writing piece but I did put it in quotation marks, is that alright?

It's good that you used quotation marks - that would probably be enough to defeat any charge of plagiarism. However, that leaves the question of referencing. You should clarify this with your institution.

Will they actually use plagiarism scans on all coursework though? There's like 4 English classes in my school with at least 20 students in each so I was wondering if they all get sent off for moderation. I also know some who did copied one sentence but did not cite, will they get heavily penalised then?

The school only has to report you for plagiarism if you have signed the candidate declaration. If you haven't signed that yet then there is time to amend your work to reference it properly.

If you were flagged, this means you have to pay special attention to those areas. You have to recheck results once again to be sure that all the quotes were cited according to your coursework requirements.
Grizwuld is right. In case you use a quote without referencing it, it is considered to be plagiarism. It is not prohibited to use quotes in your papers (unless it is stated in your requirements). Still, you have to strictly follow the citation rules.
There are several blog articles on Unicheck’s website that explain what is considered to be plagiarism. You may check them out below to understand how this software works:
https://unicheck.com/blog/prevent-plagiarism-guide/
https://unicheck.com/blog/types-of-plagiarism/

The penalties for plagiarism may be different. However, it is better not to test them as the most serious penalty is to be sent down from the course or school.
