Hi,
How does light affect the structure of chlorophyll?
And also why is a chromatogram run in the dark?

you could refer to (photosystem I and photosystem II) PSI and PSII
Chlorophyll pigments in PSI absorb light at 700nm while in PSII they absorb light at 680nm
Cyclic photo phosphorylation occurs at both PSI and PSII while non-cyclic phosphorylation occurs only at PSII

Pigments may be more visible if the chromatogram is run in the dark.
