Values for Catalase Coursework? Watch

Hi, I'm doing a biology coursework for Edexcel AS level. My coursework is on how pH affects the activity of catalase using potatoes and hydrogen peroxide.

I was wondering if anyone could suggest some appropriate values for the number of potato disks I should use? I'm using 4mm diameter x 50mm length tubes, then cutting each tube into 10mm disks. How many disks should I use for each pH value?

Also, what volumes of hydrogen peroxide would yield sufficient results?

I plan on leaving the reaction to take place for 5 minutes, should I take readings of the gas given off at intervals withing the 5 minutes, eg every 30secs?

Hi Sami,

I say to type in your exact cwk question into google and see other peoples coursework. Thats what I did for how catalase enzymes in liver are affected by temperature from an endotherm and an ectotherm.

You should use one disk for a PH value. Repeat each Ph three times and plot the average. Five minutes is too long. I did for 2 minutes and every 20 secs. 10% hydrogen peroxide work v well. 30% workd too fast for me, but you always dilute solutions! Use a gas syringe to record amount of oxygen produced. My friends did the same expt as you are going to do, but catalase enzymes from sheep liver.

Have u done your preliminary? Don't worry only need 2-3 results just to justify why u have chosen the size of the potato cylinders. My prelim i got 2 results. Worried about the actual, but luckily it all worked out! Choose different sizes of the potato cylinders and see which is best fo the reaction.

Confuzzled? just aaaask!
