Do you mean the formal address (usted) of one person:

Informal would be:
Hola. ¿Como estás?
Instead of 'Hola' you could use Buenos días or Buenas tardes.

Both the comos in your 2 questions require an accent on the first o. It’s ¿Cómo está? and ¿Cómo estás?

Whoops. And thank you (was mixing up my Spanish accented cómo with my Portuguese non-accented como!).
