An enzyme can't be used up, rather a saturation point can be reached where there is more susbstrate than enzyme, so rate of reaction remains constant as the enzyme's active sites are all being used

An enzyme is denatured when its internal bonds break and its active site changes shape, meaning it can no longer catalyse a reaction
