Question about negative control pcr test!!!

To test if your unknown isolates are yeast, you will need to set up a negative control reaction. Pick the 6 components needed for the negative control tube for PCR from the following list. One mark will be deducted for each incorrect answer:


dNTPs (tick)
Forward and reverse primers (tick)
Template DNA from your unknown isolates
RNAase A
Protease
Water (tick)
Agarose
MgCl2 (tick)
PCR buffer (tick)
Loading buffer
Glycerol
Taq polymerase (tick)
Restriction enzymes
Lysozyme


ive put tick next to what i think is correct, could someone please confirm this?

im confused.
also what is the difference between the loading buffer and the reaction buffer?
im assuming that the PCR buffer is the same as the reaction buffer.

Your negative control has everything your test reaction contains except the DNA template.
Loading buffer is for loading a gel - so if you want to run out your PCR product on an agarose gel you add loading buffer to the tube after the PCR reaction.
